Members of hypersensitive induced reaction superfamily protein involved in wheat resistance to leaf rust

Abstract

Hypersensitive Induced Reaction (HIR) proteins is from a superfamily involved in plant hypersensitive reaction (HR). In order to understand the role of members of the HIR superfamily in the wheat resistance to leaf rust pathogen, Thatcher-Lr15 line was inoculated with virulent and avirulent leaf rust races to obtain via RT-PCR full length cDNA sequences for TaHIR1-4. TaHIR2-4 were expressed in wheat young leaves, young stems and roots, while no transcripts were detected in mature seeds for all these 3 genes. Transcripts of the members of HIR superfamily in wheat leaves were differentially regulated following infection by leaf rust races as more transcripts for TaHIR2 and TaHIR3 accumulated in the resistance interaction. The expression of TaHIR2-4 proteins was up-regulated in wheat leaves inoculated with the incompatible leaf rust race as shown by Western blotting. For TaHIR2, the response in the resistance interaction was faster and stronger than that in the susceptible one. Our results demonstrate that members of wheat HIR superfamily involved in wheat resistance to infection by leaf rust pathogen at mRNA and protein levels, and provide experimental supports for further investigating the roles of HIR superfamily members during mounting of the HR defense in wheat.
